Eladia <\/strong><\/h2>\n<p>En este trabajo abordamos la caracterizacion genetica y bioquimica de una actividad carboxipeptidasica, denominada carboxipeptidasa ysps de la levadura de fision schizosaccharomyces pombe que libera leucina de sustratos peptidicos tales como el sustrato cbz-gly-leu.Hemos purificado el enzima empleando distintos pasos cromatograficos y hemos descubierto que el enzima es un oligomero. El enzima muestra gran afinidad por la leucina en el extremo carboxilo terminal, siempre que el penultimo aminoacido sea peque\u00f1o y sin carga. El valor de la km para los sustratos cbz-gly-leu, cbz-gly-gly-leu, cbz-ala-leu son de 6,7 mm, 6 mm y 20 mm, respectivamente.  la carboxipeptidasa ysps pertenece al grupo de las metaloproteasas, como se deduce del comportamiento que muestra con distintos inhibidores tipicos de proteasas y de iones metalicos.  el enzima es una glicoproteina, con tres sitios potenciales de glicosilacion, como se dedujo tras realizar distintas digestiones con endo h.  hemos obtenido anticuerpos que reconocen especificamente a la carboxipeptidasa ysps en un extracto crudo de la levadura de fision.  mediante mutagenesis con ems, se aislo una cepa deficiente en actividad carboxipeptidasica que porta con una mutacion cromosomica y recesiva, y afecta al gen estructural del enzima.  tambien comenzamos la caracterizacion molecular de la carboxipeptidasa ysps y hemos aislado plasmidos recombinantes de una genoteca que son capaces de complementar la mutacion previamente aislada.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la tesis doctoral \u00ab<strong>Analisis bioquimico, genetico y molecular de una carboxipeptidasa de la levadura de fision schizosaccharomyces pombe.<\/strong>\u00ab<\/h3>\n<ul>\n<li><strong>T\u00edtulo de la tesis:<\/strong>\u00a0 Analisis bioquimico, genetico y molecular de una carboxipeptidasa de la levadura de fision schizosaccharomyces pombe. <\/li>\n<li><strong>Autor:<\/strong>\u00a0  Arguelles Sanchez M.
