Pv: amaranthicola reconoce la secuencia gtcgac, lo cual le hace isoesquizomero de sali.  para su clonacion se construyo una genoteca de adn total de esta cepa en el vector de e.Coli pbr328. La seleccion de pu01050 se hizo en base al fenotipo de modificacion.  por medio de hibridacion y mapeo de restriccion se observo que pu01050 ademas de contener el gen que codifica para la metiltransferasa, contenia un elemento transponible de 1246 pb que se denomino is 1389. La secuencia de nt. Revelo la presencia de cuatro orfs: dos en el fragmento con actividad modificacion en la misma hebra del adn (orf1 y orf2) y dos en la secuencia de insercion en la hebra complementaria (orfa y orfb). El analisis de la secuencia de aa, permitio abscibir a orf1 el gen xamim.  la similitud mas elevada encontrada fue entre las metiltransferasas de los sistemas sali y xami, pudiendo considerar a ambas proteinas como parcialmente relacionadas desde un punto de vista evolutivo. En orf2 se detectaron dos cortas regiones que presentaban cierta similitud con la endonucleasa sali, mediante el programa blast. Por lo que orf2 podria codificar para la endonucleasa del sistema.
