En esta tesis doctoral se han evaluado las consecuencias de la mutaci\u00f3n de:  1,- la telomerasa  2,- ku86  3,- dna-pkcs y parp-1 en ratones deficientes (knock-out) para estas prote\u00ednas.  en este estudio se ha llegado a las siguientes conclusiones  1,- la actividad telomerasa no es necesaria para la inmortalidad espont\u00e1nea ni para la transformaci\u00f3n celular por la expresi\u00f3n de las oncoprote\u00ednas e1a-hrasv12, en fibroblastos embrionarios de rat\u00f3n. En ambas situaciones, la ausencia de telomerasa aumenta la inestabilidad cromos\u00f3mica y favorece la aparici\u00f3n de mecanismos alt.  2,- los fenotipos de los ratones mterc -\/- son m\u00e1s severos en ratones mterc -\/- de fondo gen\u00e9tico bl\/6, que en ratones mterc -\/- de fondo mixto bl\/6-129sv, debido a la menor longitud y heterogeneidad telom\u00e9rica del fondo bl\/6. Esto indica que el n\u00famero de generaciones de rat\u00f3n que se puede obtener en ausencia de telomerasa es proporcional a la longitud de tel\u00f3meros de la cepa.  3,- se ha comprobado que la reconstituci\u00f3n de la actividad telomerasa en ratones g4 mterc-\/- es suficiente para alargar los tel\u00f3meros cr\u00edticamente cortos, prevenir la inestabilidad cromos\u00f3mica y rescatar in vivo todos los fenotipos de los animales g4 mterc-\/-. Este estudio demuestra la aplicabilidad de la telomerasa en tratamientos de patolog\u00edas debidas a un acortamiento telom\u00e9rico cr\u00edtico.  4,- la prote\u00ednas ku86 tiene funciones esenciales para la protecci\u00f3n telom\u00e9rica.
