Un subtipo de estos enzimas son proteinas transmembrana cuya funcion es transmitir se\u00f1ales a traves de la membrana plasmatica.  en esta tesis, se presenta el trabajo realizado en la caracterizacion funcional y estructural de la proteina-tirosina-fosfatasa lar y otros miembros de la subfamilia de fosfatasas hptpd y hptps. Las fosfatasas transmembrana lar, hptpd y hptps se expresan como un polipeptido que incluye dos dominios fosfatasa intracelulares y una region extracelular formada por tres dominios inmunoglobulina y ocho dominios fibronectina. El enzima se localiza en la membrana como un complejo de dos subunidades no asociadas covalentemente, ambas derivadas de la misma proproteina por escision proteolitica. La subunidad extracelular puede ser secretada al medio. Con objeto de comprender la funcion de estas proteinas se utilizo un sistema genetico reverso para la caracterizacion de regiones funcionales necesarias para el procesamiento, la asociacion entre las subunidades y la secrecion al medio de la subunidad extracelular.  las fosfatasas lar, hptpd y hpptps estan intimamente relacionadas desde el punto de vista molecular, por lo que las formas humanas de estas proteinas fueron clonadas con el fin de realizar un estudio comparativo entre ellas a nivel estructural y de distribucion celular.  con objeto de profundizar en la funcion fisiologica de la fosfatasa lar, se buscaron proteinas que interaccionaran con la region citoplasmatica de la fosfatasa; para lo ello se utilizo un sistema genetico en levaduras, el \u00abinteraction trap assay\u00bb que permite el clonaje directo de proteinas que interaccionan con la proteina de interes.
