Tanto in vivo como in vitro el principal sustrato de fosforilacion es la subunidad c8, que se fosforiladen residuos serina. La comparacion de los fosfopeptidos obtenidos por digestion con endo glu-o a partir de la subunidad c8 fosforilada in vivo e in vitro por caseina quinasa ii muestra que los sitios de fosforilacion son similares en ambos casos. La fosforilacion in vitro de la proteinasa multicatalica no afecta a su actividad proteasa sobre proteinas o sobre peptidos fluorogenicos sinteticos.
