El residuo lys474, esta localizado en el sitio de union de atp, y la caracterizacion bioquimica de enzimas portadores de la mutacion k474r sugiere que este residuo esta implicado en la formacion del intermediario fosforilado. La lys474 es el sitio de union del analogo de la adenosina, fluoresceina isotiocianato. Para identificar regiones del enzima implicadas en la formacion del intermediar fosforilado hemos realizado un analisis de supresion intragenica de la mutacion k474r.  una de las mutaciones supresoras, v396i, es especifica de alelo y esta localizada cerca del residuo que es fosforilado durante el ciclo catalitico. Tres mutaciones supresoras, v484i, v484i\/e485k y e485k\/e486k, podrian compensar la alteracion estructural introducida por la mutacion k474r.
