En la region carboxilo de p4 existen residuos criticos para interaccionar con la rna polimerasa del huesped y activar la trancripcion, el mas importante de estos es la arginina 120. En la misma region de p4 existen aminoacidos que son importantes, como conjunto, para el mantenimiento de la curvatura inducida por p4.  estos serian r116, r118, r120 y k122. El efecto de la curvatura se manifiesta sobre los mutantes multiples. La proteina p4 se encuentra en forma de tetramero.
