El objetivo final de este trabajo ha sido profundizar en el estudio de la expresion tsh-r en distintas patolog\u00edas tiroideas humanas, asi como el factor de transcripcion especifico de tejido (ttf-1). Como modelo experimental valido para el estudio de los mecanismos moleculares que conducen al desarrollo de la neoplasia, hemos utilizado lineas celulares humanas procedentes de carcinoma papilar (npa), folicular (fro) y anaplasico (aro). Los objetivos del trabajo han sido: 1. Determinar la presencia o ausencia de expresion del tsh-r y ttf-1 en tejido retro-orbital de pacientes con enfermedad de graves-basedow. 2. Analizar la expresion del tsh-r en neoplasias tiroideas y lineas celulares procedentes de carcinoma papilar (npa), folicular (fro) y anaplasico (aro). 3. Determinar si existe o no relacion entre la expresion del tsh-r y ttf-1 en neoplasias tiroideas y 4. Estudiar la relevancia del ttf-1 en el control de la expresion del gen del tsh-r. Se realizo el analisis de la expresion del tsh-r y ttf-1 en tejido retro-orbital de pacientes con enfermedad de graves e individuos control, por la tecnica de transcripcion en reverso y reaccion en cadena de la polimerasa (rt-pcr). La expresion del tsh-r en neoplasias tiroideas y lineas celulares tumorales se estudio por ensayos de northern -blot asi como por la tecnica de rt-pcr. Para intentar determinar la importancia del factor de transcripcion ttf-1 en el control de la expresion del tsh-r, utilizamos los ensayos de retardo de banda en gel y transfecciones celulares transitorias. Los resultados han sido los siguientes: 1. Encontramos expresion del tsh-r en tejido retro-orbital de pacientes con enfermedad de graves, sugiriendo una mayor expresion<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la tesis doctoral
