Bel\u00e9n L\u00f3pez Brito <\/strong><\/h2>\n<p>Las hidrogenasas (ni-fe) de bacterias endosimbioticas de leguminosas (rhizobium y bradyhizobium) son metaloenzimas constituidas por una proteina heterodimerica que contiene niquel en su centro activo y grupos fe-s en numero variable. Estas hidrogenasas funcionan oxidando el h2 generado por la nitrogenasa durante la reduccion de n2 en los nodulos. Este sistema de oxidacion de hidrogeno aumenta la eficiencia energetica de la simbiosis y constituye una fuente potencial de mejora de la fijacion de nitrogeno y de la productividad de las leguminosas.  dentro de un objetivo general centrado en el analisis del sistema de oxidacion de hidrogeno de r. Leguminosarum, en esta tesis se abordan tres objetivos concretos: i) la evaluacion de los requerimientos y analisis del efecto del niquel en la sintesis y actividad hidrogenasa de r.  leguminosarum bv. Viciae en simbiosis con guisantes; ii) el analisis de la regulacion de la expresion simbiotica de los genes estructurales de la hidrogenasa de r.  leguminosarum bv. Viciae, iii) la busqueda de genes reguladores en la region de dna contigua al promotor de los genes hupsl. Los resultados mas relevantes han sido los siguientes:  la actividad hidrogenasa de r. Leguminosarum bv. Viciae en nodulos de guisantes esta limitada por la disponibilidad de niquel para la planta en el suelo o en soluciones nutritivas. En condiciones de deficiencia de niquel, las proteinas estructurales de la hidrogenasa no se procesan y la enzima se acumula en una forma inmadura e inactiva. El procesamiento requiere, ademas de niquel, los productos geneticos derivados de los genes hup y hyp accesorios.  el promotor de los genes estructurales de la hidrogenasa (phup sl) presenta una estructura del tipo de promotores -12\/-24 caracteristica de genes-nif. Mediante ensayos de delecion se identifico una region localizada entre las posiciones -173\/-88, esencial para la actividad de este promotor en nodulos de guisantes.
