Consiste en la transfeccion transitoria de adipocitos marrones diferenciados en cultivo, a partir de celulas precursoras obtenidas de depositos de tejido adiposo marron de raton.  2.- El estudio de la actividad de la construccion (-4551)ucp-cat, que contiene 4,5 kb de la region 5\u00c2\u00bfdel gen ucp dirigiendo el gen reporter cat, indica una expresion preferencial en los adipocitos marrones respecto a otros tipos celulares heterologos como hepg2 y cho. Dicha expresi\u00f3n preferencial se mantiene en la construccion (-896) ucp-cat, dirigida por la region proximal entre -896\/+92.  3.- Hemos establecido un mapa de los lugares deuni\u00f3n a proteinas nucleares en la region proximal del gen ucp mediante analisis \u00abfootprint\u00bb con dnasal. se han identificado dos lugares: lugar l (-402\/-361) y lugar b(-512\/-487) con union diferencial a proteinas nucleares de tejido adiposo marron respecto higado.  4.- Los factores de transcripcion c\/ebpa y b son activadores de la transcripcion del gen ucp. Dicha acci\u00f3n es mediada principalmente por dos lugares de la region proximal del gen que unen dichas proteinas (-457\/-440 y -335\/-318) y que son capaces de conferir respuesta a c\/ebp a promotores heter\u00f3logos.  5.-La expresion de (-4551) ucp-cat se activa por noradrenalina, ampc y sobre-expresion de la subunidad catalitica de la proteina kinasa a. Dicho efecto es independiente de los elementos de regulacion tejido-especifica y requiere principalmente la region proximal -157\/-54 del gen ucp.  6.-Dicha region contiene un elemento (-139\/-122) capaz de unir creb y proteinas relacionadas con creb presentes en nucleo de tejido adiposo marron. C-jun esuna de las proteinas nucleares de tejido adiposo marron que unen icho lugar.  7.- C-jun actua como represor de la expresion basal y del estimulo por proteina kinasa a de (-4551) ucp-cat.
