Yolanda <\/strong><\/h2>\n<p>El gen pin0 se ha localizado en la kb 3475 del mapa fisico de rudd. Los analisis de restriccion llevados a cabo a\u00f1aden modificaciones al mapa de rudd, de gran utilidad para los estudios que se lleven a cabo en esta zona del cromosoma. El gen pin0 se ha clonado en plasmidos con distinto numero de copias, pero no se ha podido estudiar el efecto del aumento de la expresion de pin0 por la ausencia en el gen clonado del codon de iniciacion. El gen pin0 se ha mutado y el analisis de los parametros del ciclo y replicacion ha permitido concluir que el gen pin0 es dispensable y que su producto no es la proteina carente de isoleucina necesaria para el inicio de replicacion en e. Coli propuesta por guzman et al.  (res. Microbiol. 142:137-140, 1991). La secuenciacion del gen pin0 ha permitido analizar los elementos reguladores que comprenden su region promotora. En este analisis se ha identificado un promotor principal de tipo restringido y dos promotores secundarios, uno de los cuales estaria regulado por choque termico. Al analizar la secuencia se ha detectado la ausencia de 15 nucleotidos que incluyen el codon de inicio de pin0. La presencia de este quincemero en la region promotora de e.
