Dolores <\/strong><\/h2>\n<p>Tomando en consideracion la naturaleza proteica de numerosas nuevas moleculas terapeuticas y antigenicas y la necesidad apremiante de desarrollar formas adecuadas para su administracion, el objetivo global de la tesis doctoral se centro en el desarrollo microsferas y nanosferas biodegradables, destinadas a la administracion parenteral, capaces de liberar de un modo controlado las proteina que llevan encapsuladas. Como polimeros biodegradables formadores de las micro y nanosferas se eligieron los copolimeros del acido lactico y glicolico (plga) debido a su probada aceptabilidad para la administracion a humanos.  el objetivo inicial del trabajo consistio en la busqueda del mejor procedimiento de microencapsulacion que permita encapsular, de manera eficaz e integra, este tipo de macromoleculas. Para ello, se ensayaron dos procedimientos de microencapsulacion (extraccion\/evaporacion del disolvente en fase externa acuosa y oleosa) y se seleccionaron dos proteinas modelo de diferente punto isoelectrico y peso molecular (seroalbumina bovina y lisozima de huevo). Ademas, se estudio la influencia del peso molecular del plga y de la incorporacion o no de dos agentes estabilizantes de las proteinas, sobre el tama\u00f1o de las particulas y sobre la capacidad de las mismas para la encapsulacion y liberacion controlada de proteinas. Aprovechando la gran versatilidad que ofrece el procedimiento de extracion del disolvente en fase externa acuosa, fue posible reducir el tama\u00f1o de las particulas hasta valores submicrometricos, de tal forma que se han podido desarrollar, por primera vez, nanosferas de plga que encapsulan eficazmente proteinas.
