Estudiar a nivel molecular y celular las acciones de agentes citotoxicos de interes biologico 2. Comprobar el papel de los genes p53 y p16 como determinantes de los efectos biologicos que se desencadenan tras tratamiento con radiacion gamma o hipertermia 3. Analizar los aspectos metodologicos relativos al estudio de la apoptosis.  los modelos tumorales utilizados son los siguientes:  mcf-7 (bus, bb, gs), t47d, evsa-t, mda mb 231, rt112, d283med, hl60, l929, ito, mrc-5.  entre las tecnicas aplicadas destacan: determinacion inmunohistoquimica de p16 y p53, northern blotting, citometria de flujo, determinacion del nivel de acido lactico extracelular, western blotting, extraccion y electroferesis del dna fragmentado, tecnica comet, end-labelling o tunel, determinacion del pico sub-g0.  entre las conclusiones obtenidas destacan las siguientes:  1. El tratamiento de las lineas celulares con dosis de 6 gy de radiacion refleja la existencia de modificaciones en la progresion de las celulas a lo largo del ciclo celular. En todos los modelos utilizados se encontro parada en la fase g2\/m del ciclo celular mientras que la detencion en la fase g1 se aprecia solo en aquellas celulas con niveles constitutivos de p53 bajos.  2. En las mismas condiciones experimentales anteriores se demostro el incremento de los niveles intracelulares de p53 solo en aquellas celulas en las que se produce la parada en la fase g1.  3. Se ha encontrado una relacion consistente entre sf2, y la funcionalidad de la proteina.  4. El analisis combinado de p53 y p16 demostro que: a) los niveles constitutivos de ambas proteinas son independientes entre si b) los cambios en los niveles intracelulares de p16 tras irradiacion con 6 gy no se relacionan con la radiosensibilidad celular intrinseca c) la hipertermia origina el incremento intracelular de la proteina p5<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la tesis
