Gabriela <\/strong><\/h2>\n<p>El trabajo realizado en esta tesis se ha centrado en la caracterizacion fisica y funcional de los origenes de replicacion de la cadena retrasada (ssoa y ssou) del plasmido de amplio espectro de huesped pmv158. En primer lugar, se acoto in vivo la funcion del ssoa, en un fragmento de 199 pb del plasmido y se comprobo su actividad en diferentes huespedes. Del mismo modo, se establecio la funcion del ssou, en varios hospedadores bacterianos. Se determinaron los puntos desde donde se inicia la sintesis de la cadena retrasada a partir de ambos origenes de replicacion y se analizo la influencia de mutaciones en las regiones conservadas del ssoa en la eficiencia de replicacion (in vivo e in vitro) y especificidad de inicio de esta cadena (in vitro). Para ello, se puso a punto un sistema de replicacion que utiliza extractos acelulares de pneumococos, asi como tambien se consiguieron las condiciones necesarias para la replicacion a partir de estos origenes en el sistema de s. Aureus. Se pudieron determinar algunos de los factores del huesped implicados en la conversion de ss- dsdna plasmidico y al nivel en que estos actuan. Se ideo un sistema de transcripcion in vitro, que permitio la sintesis de rna cebadores a partir de sustratos de dna monocatenario del plasmido pmv158 que contenian la se\u00f1al ssoa o derivadas de esta.
