La comprobacion de una efectiva incorporacion de dnc a las histonas, y la subsiguiente determinacion de las posiciones de modificacion ha permitido evaluar la utilidad de los diferentes derivados. Una vez determinada la capacidad de las histonas como sustratos de tgasa, dos estrategias se han seguido: el empleo de los derivados fluorescentes en experimentos de reconstitucion, con el objeto de analizar cambios en los parametros fluorescentes de la sonda a fin de estudiar cambios conformacionales en el nucleosoma. La segunda estrategia es el analisis de cambios en la reactividad de las glutaminas de las histonas como una manera de seguir cambios en la estructura del nucleosoma. Esta segunda via constituye una novedosa estrategia, no solo dentro del campo de la cromatina, sino dentro del campo de la transglutaminasa.
