Dolores <\/strong><\/h2>\n<p>La purificacion y caracterizacion de la alfa-amilasa de s. Griseus imru 3570 dio como resultado la aparicion de dos proteinas con actividad amilasa una de 60 kda y otra de 49 kda, demostrandose la existencia de un procesamiento post-traduccional. Los estudios de optimizacion se realizaron modificando la fuente de carbono, siendo el medio mas productivo lechevalier con 1% de maltosa. Otra tecnica para incrementar la produccion fue la sustitucion del promotor amy por el promotor saf, lo cual produjo un incremento de 15,6 veces en actividad y de 10,4 veces en cantidad de proteinas. Se realizaron estudios que demostraron que no se producia acumulacion de arnm intracelularmente, por lo tanto esta no es una etapa limitante para la secrecion. El estudio de las fusiones traduccionales alfa-amilasa y el gen hcrf puso de manifiesto varios hechos: i) al recortar la proteina portadora disminuye la produccion de proteinas secretada; ii) al fusionar el gen hcrf a la proteina portadora aun disminuye mas la produccion; iii) el cambio de pamy por psaf incrementa la produccion.
