El oligotipado consiste en la deteccion de mutaciones puntuales en los genes estructurales de las b-lactamasas tem mediante la hibridacion con sondas de oligonucleotidos, obteniendo un perfil de hibridacion que constituye el oligotipo. El objetivo del presente trabajo fue la puesta a punto de tecnicas de biolog\u00eda molecular y su aplicacion al estudio de las bases geneticas de la resistencia a cefalosporinas de 3 generacion mediada por beta-lactamasas de espectro ampliado tem-derivadas. Se estudiaron 28 cepas de enterobacterias portadoras de genes blatem mediante determinacion de su fenotipo de resistencia, isoelectroenfoque analitico, analisis plasmidico y oligotipado. Se detectaron 5 oligotipos diferentes, que se correspondian con los genes productores de beta-lactamasas tem-1, tem-2, tem-4, tem-6 y tem-10. De las 28 cepas estudiadas, 4 eran productoras de beta-lactamasas de espectro ampliado tem-derivadas, estando sus genes blatem localizados en plasmidos de alto peso molecular. El fenotipo de resistencia y el ief fueron insuficientes para caracterizar estos enzimas. Por el contrario, la hibridacion con sondas de oligonucleotidos no radiactivas, en combinacion con la obtencion de adn diana mediante pcr, permitio la caracterizacion especifica de las beta-
