La solubilizacion optima de la actividad se puede llevar a cabo mediante dos procedimientos alternativos:  ultasonacion o tratamiento de las membranas con el detergente no ionico n-octil beta-d-glucopiranosido (og).  la ultasonacion permite la soluilizacion de la actividad ceh presente en 100mg de proteina microsomal en menos de 8 min recuperando el 95% de la actividad ceh en el 34% de la proteina. El 100% de la actividad ceh asociada al 42% de las proteinas se solubiliza tambien tratando los microsomas con el detergente og a concentracion 30 mm manteniendo en 4 la proporcion en peso entre detergente y proteina.  la actividad ceh de microsomas de higado de rata se purifica en las siguientes etapas: separacion de membranas microsomales por fraccionamiento subcelular, lavado de membranas, solubilizacion por ultrasonacion, precipitacion al 7% (p\/v) de peg, aclaramiento con cristales de hidroxiapatitoal al 0,1% (p\/v), cromatografia de intercambio ionico en columnas hitrap q y cromografia en hidroxiapatito. El poder mas resolutivo de esta purificacion recae sobre la etapa de intercambio ionico, gracias a la utilizacion de un inusual proceso de elucion, en el que se aplican escalones de fuerza ionica creciente partiendo en cada caso desde el 0% del eluyente salino. Dada la marcada inestabilidad de la muestra purificada, el rendimiento era del 3,21% y el factor de purificacion de 12,4 veces y representanba el 0,25% de la proteina microsomal de partida.
