Se utilizan 250 ratas macho wistar. Se mide la ingesta de agua durante 2 horas retirando los alimentos. Se realiza una curva dosis-respuesta de tolbutamida observando un efecto dipsogenico entre 60 y 200 mg\/kg p.C.  para posteriores experimentos se utilizan 80 mg de tolbutamida i.P.\/Kg p.C. Y 40 mg de acetohexamida i.P.\/Kg p.C. Con dichas dosis la glucemia disminuye significativamente.  no existe relacion entre la sed inducida por ambos aho y cambios volumetricos ni osmolares. La nefrectomia y adrenalectomia no modifican la accion de estas drogas sobre la ingesta de agua. Tampoco esta relacionada con mecanismos histaminergicos puesto que el bloqueo de los receptores de este sistema no provoca cambios significativos en dicha ingesta. El captopril aumenta la accion dipsogenica de estas drogas, lo que sugiere que el sistema renina-angiotensina esta involucrado en este fenomeno. La sed inducida por sulfonilureas esta mediada por el sistema beta-adrenergico cuyo bloqueo con propranolol disminuye la ingesta liquida. La administracion de tolbutamida en ratas diabeticas no disminuye la ingesta de agua. Las biguanidas i.P. En dosis de 50 a 600 mg\/kg p.C.
