Pilw contiene 12 genes: 10 genes para la produccion de pili que denominamos trwd, e, f, g, h, i, j, k, l y m, un gen kor y un gen de exclusion de superficie que llamamos eex.  trwh es ademas un gen kil.  la transcripcion de los genes en pilw se realiza en tres transcritos diferentes, que denominamos i, ii y iii. La expresion in vivo por el sistema de minicelulas muestra proteinas de peso molecular 40, 47, 9, 90, 7 y 11 kda expresadas por los genes trwd, e, eex, trwk, trwl y kor, respectivamente. La proteina trwl es la proteina estructural del w-pilus, y aparece en preparaciones de pili en forma procesada. Las proteinas de la region pilw presentan similitud de secuencias de aas con las proteinas del operon virb de plasmidos ti de agrobacterium tumefaciens. Proteinas similares en los dos sistemas comparten motivos funcionales contenidos en sus secuencias de aas.
