Luisa <\/strong><\/h2>\n<p>Se ha secuenciado la region del plasmido r388 implicada en el metabolismo conjugativo del dna (mobw), que ocupa un segmento de dna de 5.163 pb. Mobw consiste en el origen de transferencia (orit) y los genes trwa (366 pb), trwb (1.524 pb) y trwc (2.901 pb).  orit se ha delimitado en un segmento intergenico de 330 pb. Destacan en el un tandem de repeticiones directas, cuatro repeticiones invertidas, un alto contenido en a+t y un segmento con curvatura intrinseca. Funcionalmente se distinguen tres dominios en orit: dos en los extremos, requeridos para una alta frecuencia de movilizacion, y un dominio central esencial de 60 pb que es suficiente para la funcion orit. La organizacion de orit es similar a la del orit del plasmido incn r46, presentando ambas secuencias un 53% de homolog\u00eda.  trwa, trwb y trwc se transcriben en una misma direccion. Los datos de la secuencia sugieren que formen una unidad transcripcional.  los niveles de expresion son muy bajos debido a la ausencia de promotores fuertes. Los productos de estos genes son proteinas de 13.402 da (trwa), 56.280 da (trwb) y 107.550 da (trwc). Las tres proteinas han sido superproducidas y presentan tama\u00f1os aparentes de 15, 60 y 105 kda, respectivamente.  trwa presenta una cremallera de leucinas. Trwa es homologa a traj de rp4 y r751 (incp) y a nika de r64 (inci), todas ellas implicadas en la reaccion inicial de rotura en orit.  trwb es una proteina asociada a membrana interna. Presenta una posible secuencia de union para atp. Su secuencia es homologa a la de las proteinas trad de f y r100 (incf), presumiblemente implicadas en el transporte conjugativo del dna hacia el exterior celular.  la region amino-terminal de trwc es la unica region de mobw necesaria para que ocurra recombinacion especifica entre dos copias de orit.  esta region es homologa a la correspondiente de la proteina trai de f, que contiene la actividad de endonucleasa de orit.
