Jose <\/strong><\/h2>\n<p>La cebada es una de las materias primas en la elaboracion de la cerveza. La pared de las celulas del endospermo amilaceo esta constituida mayoritariamente por un polisacarido complejo: beta-(1,3)-(1,4)-glucano. Este polimero causa diferentes problemas a lo largo de la elaboracion de la cerveza.  este trabajo se centra en el clonaje y caracterizacion en e. Coli de un gen que codifica una beta-(1,3)-(1,4)-glucanasa de bacillus polymyxa que degrade este polimero. Esta especie se selecciono entre 24 especies del genero bacillus. A partir de una genoteca de b. Polymyxa en e. Coli se aislaron 2 clones con actividad beta-(1,3)-(1,4)-glucanasa, denominados bpl1000 y bpl5800. Este ultimo clon degrada tambien el xilano, al igual que otro clon aislado de la misma genoteca, denominado xynl.  a partir de los mapas fisicos de los plasmidos aislados de estos clones y experimentos de hibridacion southern se deduce que los genes responsables de la actividad xilanasa y glucanasa se localizan adyacentes en el genoma de b. Polymyxa. La caracterizacion de la endoglucanasa en e. Coli se abordo con su localizacion y determinacion de su peso molecular.  el estudio del gen abarco su localizacion en el fragmento clonado y la determinacion de su orientacion mediante experimentos de subclonajes. Tambien se realizo el clonaje y expresion del gen en b. Subtilis. Esta caracterizacion se completo con la secuenciacion de ambos genes y con un estudio de homolog\u00eda. Finalmente, se llevo a cabo el analisis de la transcripcion de los genes citados en b.
