Tambien ha sido realizado un exhaustivo estudio fisico-quimico de la interaccion entre las histonas y dna monocadena, incluyendo un detallado analisis del efecto de las histonas sobre la reaccion de renaturalizacion de cadenas de dna complementarias. Esta segunda parte de la tesis ha requerido el clonaje de fragmentos de dna de origen nucleosomal y el posterior dise\u00f1o de un metodo de obtencion en cantidad suficiente de las dos cadenas complementarias de los fragmentos de dna obtenidos. Ademas de los metodos clasicos de extraccion de dna plasmidico y virico y de extraccion de histonas, que incluyen gradientes de densidad, cromatografias de intercambio ionico y tecnicas microbiologicas, tambien han sido usadas en el estudio de la interaccion histona-dna monocadena las electroforesis en geles de agarosa y de poliacrilamida, asi como digestiones con diversas nucleasas
