El objetivo de este trabajo fue establecer una tecnica de obtencion de islotes humanos como sustrato de estudios inmunologicos y futuro material transplantable. Para ello se compararon una tecnica manual y una automatizada (10 y 35 pancreas respectivamente). Los islotes obtenidos fueron el sustrato de estudios de: 1) expresion y modulacion de las moleculas de adhesion icam-1 y lfa-3 en celulas beta, 2) localizacion del autoantigeno glutamato descarboxilasa (gad), 3) reactividad antimembrana de los autoanticuerpos de pacientes diabeticos empleando la tecnica de inmunofluorescencia indirecta. La tecnica automatizada fue superior a la manuel en terminos de rendimiento, viabilidad y pureza de los islotes obtenidos ofreciendo islotes aptos para el transplante tal como indica la secrecion insulinica in vitro. La expresion de moleculas de adhesion fue negativa en condiciones frescas pero aparecia en los islotes en cultivo y mas aun bajo la influencia de linfoquinas (ifn-gamma, tnf-alfa). El autoantigeno gad se localizo no solo en celulas beta sino tambien en las alfa. Finalmente no se hallo reactividad de los autoanticuerpos con celulas insulares viables (en condiciones basales o sometidas a estres) indicando que el autoanigeno no se encuentra en la membrana de las celulas beta.
