Coli es esencial para la septacion. Su gen (pbpb) se localiza en un extremo del culster de morfogenes del minuto 2 del cormosoma. Durante la tesis se han secuenciado 192 kb previas a los genes orfa y pbpb, habiendose identificado dos nuevas orfs, que hemos denominado orfc y orfb. Esta ultima codifica para una proteina de 35000 d, que ha sido detectada en un sistema de maxicelulas. Se ha determinado, determinado, mediante ensayos de complementacion y de mapeo con s1, la existencia de promotores independientes para orfc, orfb, orfa y pbpb, aunque posiblemente se sintetizan transcritos que abarcan mas de una orf. La actividad de los promotores de orfb esta repimida por lexa.. Se ha detectado tambien la produccion de un rna antisentido que podria regular la expresion de orfa. Se han caracterizado 14 formas alteradas de la pbp3, obtenidas por mutagenesis dirigida, con cambios en regiones del extremo amino y carboxilo importantes para la localizacion y exportacion de la pbp3. Su analisis ha demostrado que la pbp3 no puede ser procesada por la peptidasa se\u00f1al ii, que la pbp3 se exporta por un mecanismo dependiente de seca y secy y que en la region carboxilo terminal pueden definirse diferentes regiones implicadas en el procesamiento proteolitico del extremo carboxilo, en la funcionalidad de la proteina y en su topolog\u00eda.
