{"id":14237,"date":"2001-03-12T00:00:00","date_gmt":"2001-03-12T00:00:00","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/receptores-p2x-en-neuronas-de-purkinje-de-cerebelo-de-rata-neonatal\/"},"modified":"2001-03-12T00:00:00","modified_gmt":"2001-03-12T00:00:00","slug":"receptores-p2x-en-neuronas-de-purkinje-de-cerebelo-de-rata-neonatal","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/quimica\/receptores-p2x-en-neuronas-de-purkinje-de-cerebelo-de-rata-neonatal\/","title":{"rendered":"Receptores p2x en neuronas de purkinje de cerebelo de rata neonatal"},"content":{"rendered":"<h2>Tesis doctoral de <strong>  Garcia Martinez De Lecea Marta <\/strong><\/h2>\n<p>Las neuronas de purkinje han sido y son centro de gran inter\u00e9s debido a sus exclusivas caracter\u00edsticas electrofisiologicas y a su importante papel como la \u00fanica v\u00eda de salida de informaci\u00f3n de la corteza cerebelosa. considerando que la mayoria de los datos sobre la expresi\u00f3n de mrna de las subunidades p2x en cerebelo proceden de ratas adultas, decidimos establecer la edad del cultivo en una semana despues del nacimiento del animal (p7), momento en que las neuronas se encuentran en una etapa m\u00e1s avanzada del desarrollo y de ese modo trabajar con neuronas que presenten, en la medida de lo posible, un comportamiento y unas propiedades m\u00e1s proximas a las de las neuronas de purkinje adultas. Gracias al empleo de tecnicas de electrofisiolog\u00eda, fluorimetria y biolog\u00eda molecular, hemos puesto de manifiesto la presencia de receptores nucleot\u00eddicos p2x, en cerebelo. En cultivos primarios de cerebelo de p7 se expresan mrnas para todas las subunidades p2x, salvo la subunidad p2x5. Las subunidades p2x1,p2x2, p2x3, p2x4 y p2x6 se localizan especificamente en las neuronas de purkinje y forman canales funcionales de estequiometria aun no caracterizada molecularmente. Dichos receptores ionotr\u00f3picos p2x son muy permeables a ca2+ y pueden inducir un aumento significativo de [ca2+]i incluso sin el concurso de los canales de ca2+ dependientes de voltaje, por entrada de ca2+ a trav\u00e9s del propio canal p2x. En el estadio p7 existe una poblaci\u00f3n mayoritaria y dominante de receptores p2x2\/6, sensibles al antagonista ppads y potenciados por zn2+ y acidificaci\u00f3n extracelular que corresponde a la formaci\u00f3n de receptores heterom\u00e9ricos por las subunidades p2x6 y las p2x2 y p2x2b, distinguibles por su diferente cin\u00e9tica de desensibilizaci\u00f3n. La expresi\u00f3n de las subunidades p2x cambia a lo largo del desarrollo el cerebelo. Se producen una sustituci\u00f3n durante la maduraci\u00f3n de subunidades p2x2 por p2x4, cuya expresi\u00f3n relativa cambia 90 veces de p1 al es<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la
