{"id":14379,"date":"2001-11-12T00:00:00","date_gmt":"2001-11-12T00:00:00","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/rediseno-de-la-eficiencia-catala%c2%adtica-y-de-la-termorresistencia-de-la-beta-i-3i-4glucanasa-de-bacillus-licheniformis\/"},"modified":"2001-11-12T00:00:00","modified_gmt":"2001-11-12T00:00:00","slug":"rediseno-de-la-eficiencia-catala%c2%adtica-y-de-la-termorresistencia-de-la-beta-i-3i-4glucanasa-de-bacillus-licheniformis","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/quimica\/rediseno-de-la-eficiencia-catala%c2%adtica-y-de-la-termorresistencia-de-la-beta-i-3i-4glucanasa-de-bacillus-licheniformis\/","title":{"rendered":"Redise\u00f1o de la eficiencia catal\u00edtica y de la termorresistencia de la beta (i-3)(i-4)glucanasa de bacillus licheniformis"},"content":{"rendered":"<h2>Tesis doctoral de <strong> Juan  Antonio Cedano Rodr\u00edguez <\/strong><\/h2>\n<p>Introducci\u00f3n  las beta(l-3)(l-4) glucanasas son enzimas de gran inter\u00e9s biotecn\u00f3logico tanto en la industria cervezera como en la de fabricaci\u00f3n de piensos.  los objetivos de esta tesis han sido: redise\u00f1ar la glucanasa de bacillus licheniformis con objeto de tratar de mejorar la actividad enzim\u00e1tica y determinar la funci\u00f3n del lazo mayor del centro activo en dicho proceso, as\u00ed como analizar y redise\u00f1ar la estabilidad t\u00e9rmica del enzima, en particular determinar la influencia del lazo mayor en la desnaturalizaci\u00f3n t\u00e9rmica, relacion\u00e1ndolo con el proceso y tipo de plegamiento.  an\u00e1lisis de los mutantes de saturaci\u00f3n en la posici\u00f3n 58 de glucanasas  1,- la reducci\u00f3n de volumen de la cadena lateral aumenta la actividad, aumentando la constante catal\u00edtica kcat, si bien disminuye la km. El mutante m58g presenta la actividad m\u00e1s alta, hasta 6 veces m\u00e1s que el silvestre, y elmutante m58a presenta m\u00e1s de 4 veces la actividad del silvestre, ambos, en t\u00e9rminos de kcat\/km.  2,- los residuos hidr\u00f3filos disminuyen la actividad.  3,- unicamente en mutantes arom\u00e1ticos como m58f y m58w, se aprecia una disminuci\u00f3n de valores de km, respecto al enzima silvestre.  el an\u00e1lisis de la termorresistencia  1,- con respecto al medio, ser\u00edan claves el ph (por p\u00e9rdida de interacciones ph dependientes), el tamp\u00f3n (por su posible papel quelante), y la presencia de sustrato (porque estabilizar\u00eda).  2,- con respecto a la estructura ser\u00edan claves los amino\u00e1cidos del lazo y la coordinaci\u00f3n del ca++. La coordinaci\u00f3n del ca++ puede ser mejorada introduciendo una nueva interacci\u00f3n entre el cati\u00f3n y la beta 15, como en el caso del mutante n236d.  3,- finalmente, la termoinactivaci\u00f3n de la glucanasa de bacillus licheniformis, en todos los mutantes estudiados, siguen un patr\u00f3n bif\u00e1sico. Su an\u00e1lisis cin\u00e9tico permite justificar el efecto de diversos mutantes y el comportamiento del enzima frente a la temperatura.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la tesis
