Y daban, j. R. (1988) j. Mol. Biol. 204, 141-154). Los experimentos de dispersion de rayos x de sincrotron y modelaje realizados sobre los complejos dna-histona en disolucion a diferentes fuerzas ionicas (entre muy baja fuerza ionica y 0.2 m naci) indican que las histonas h2a,h2b no pueden plegar el dna, sino que producen estructuras alargadas. Los complejos reconstituidos preparados a concentracion salina fisiologica tanto a partir de las cuatro histonas de la particula nucleo como a partir de las histonas h3,h4 en ausencia de las h2a,h2b, son estructuras completamente plegadas con un radio de giro similar al correspondiente a las particulas nucleo nativas (42 a). El conjunto de los estudios realizados sugiere que la conformacion abierta de los complejos dna-(h2a,h2b) facilita la implicacion de dicha estructura como intermediario transitorio en la reaccion de formacion del nucleosoma a fuerza ionica fisiologica.  a pesar del extenso uso de la tecnica de electroforesis de poliacrilamida-sodio dodecil sulfato (page-sds) en quimica de proteinas, la estructura de los complejos proteina-sds esta todavia en entredicho.  el modelo mas considerado actualmente asume que dichas estructuras consisten en bastones rigidos con diametro constante y longitud en funcion del peso molecular. En contraste, los estudios de dispersion de rayos x de sincrotron, modelaje y crio-microscopia electronica realizados en esta tesis indican que la estructura de los complejos proteina-sds consiste en una sucesion de micelas mixtas proteina-sds (en numero proporcional al peso molecular de la proteina) unidas por segmentos de cadena polipeptidica sin sds. Dichas micelas tienen una estructura esferica con un radio de 17 a, y tienen una distribucion radial de densidad electronica heterogenea en la cual el<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos
