Los determinantes de la baja actividad de los proenzimas no parecen relacionarse con estados de ionizacion diferentes con respecto al enzima activo (los pk de grupos funcionales cataliticamente criticos son muy parecidos), sino mas bien con residuos de la proteina correspondientes a los subsitios de reconocimiento secundario de sustratos, que establecen contactos directos con residuos del segmento de activacion en los proenzimas. Por esta razon, se produce una falta de interacciones o contactos anomalos que impiden tanto la union de inhibidores anionicos como que los sustratos puedan colocarse en el centro activo correctamente
