Teresa <\/strong><\/h2>\n<p>La finalidad de esta tesis ha sido el estudio del papel que juegan las gtpasas de la familia rho en la bios\u00edntesis de la pared de schizosaccharomyces pombe. Estas prote\u00ednas est\u00e1n conservadas evolutivamente y son necesarias para la organizaci\u00f3n del citoesqueleto de actina que, a su vez, es esencial para la bios\u00edntesis de la pared celular. Hemos estudiado la ruta de se\u00f1alizaci\u00f3n de rho1p y rho2p, y su interacci\u00f3n con otras prote\u00ednas implicadas en la bios\u00edntesis de la pared celular de s.Pombe como las quinasas pck1p y pck2p.  las prote\u00ednas de la familia rho alternan entre una forma activa. Cuando est\u00e1n unidas a gtp, y una forma inactiva, cuando est\u00e1n unidas a gdp. La transici\u00f3n entre una conformaci\u00f3n u otra es negativamente modulada por prote\u00ednas activadoras de la capacidad gtp\u00e1sica (gaps), que favorecen la hidr\u00f3lisis del gtp a gdp, y juegan un papel fundamental en la especificidad de las funciones de rho. Tambi\u00e9n hemos descrito en este trabajo el gen denominado gpr1+ que codifica una prote\u00edna gap de rho1p. La sobreexpresi\u00f3n de gpr1+ causa una lisis celular r\u00e1pida, y una disminuci\u00f3n del 40% de la actividad (1,3)beta-d gluc\u00e1n sintasa, respecto de una estirpe silvestre. por el contrario, c\u00e9lulas carentes del gen gpr1+ son viables y muestran un ligero fenotipo morfol\u00f3gico a 37\u00c2\u00ba. La actividad (1,3)beta-d gluc\u00e1n sintasa de estas c\u00e9lulas est\u00e1 considerablemente incrementada.  la sobreexpresi\u00f3n de rho1+ en c\u00e9lulas gpr1 es letal y cuasa un incremento de la actividad (1,3)beta-d-gluc\u00e1n sintasa mayor que el detectado en c\u00e9lulas silvestres que sobreexpresan rho1+. Gpr1p co-inmunoprecipita con rho1p, y muestra actividad espec\u00edfica gap sobre rho1p tanto in vitro como in vivo. grp1p participa en la regulaci\u00f3n de la estabilidad de las prote\u00ednas quinasas pck1p y pck2p mediada por rho1p.
