En primer lugar, un analisis preliminar de la fraccion de dna repetitivo del genoma haploide, encontrandose que el enzima apai libera 3 secuencias repetitivas diferentes en frecuencia, secuencia y organizacion genomica. Ademas, se ha caracterizado el gen de la proteina 01 mediante la construccion de librerias de expresion, obteniendose su cdna del que se ha deducido la secuencia aminoacida estableciendose indices de similitud con proteinas conocidas y determinando algunos motivos estructurales en su secuencia. Ademas, el cdna ha sido utilizado para comprobar que el gen es de copia unica y para aislar clones genomicos, de una genoteca de dna espermatico. Los clones genomicos aislados pusieron de manifiesto que el gen de la proteina 01 es discontinuo y con al menos dos intrones. Finalmente, se ha estudiado el efecto de la proteina 01 en la reconstitucion de cromatina in vitro, utilizando extractos acelulares de oocito de xenopus laevis observandose que las nucleoproteinas reconstituidas en presencia de 01 mantienen las caracteristicas hidrodinamicas, la topolog\u00eda del dna y los constituyentes proteinicos de la cromatina control o canonica. Esto se debe a que una proteina mayoritaria en el oocito, denominada nucleoplasmina, es capaz, en ausencia de otros elementos, de disociar complejos altamente compactados de dna-01 formados a una
