Se ha expresado la topoisomerasa i de e. Coli que selectivamente relaja la supehelicidad negativa del dna, en las c\u00e9lulas deficientes en actividad topoisomerasa y se ha estudiado el efecto de esta actividad en la estabilidad de los minicromosomas y genes ribos\u00f3micos.  se ha fusionado las dos subunidades que codifican para la dna gyrasa de e.Coli y se ha expresado esta prote\u00edna en s.Cerevisiae. Este enzima relaja selectivamente la superhelicidad positiva. Se ha estudiado es efecto de esta actividad es la estabilidad de los minicromosomas y de los genes ribos\u00f3micos.  los resultados indican que un d\u00e9ficit de actividad relajadora del dna provoca que minicromosomas circulares de peque\u00f1o tama\u00f1o generen amplias distribuciones de mult\u00edmeros las cuales se encuentran en equilibrio din\u00e1mico alrededor de formas circulares de tama\u00f1o superior.  la frecuencia de recombinaci\u00f3n mit\u00f3tica hom\u00f3loga de estas c\u00e9lulas se encuentra incrementada en dos \u00f3rdenes de magnitud.  la relajaci\u00f3n selectiva de la superhelicidad positiva, mejora su velocidad de crecimiento, evita la escisi\u00f3n de los genes ribos\u00f3micos, evita la multimerizaci\u00f3n de minicromosomas y la hiper-recombinaci\u00f3n mit\u00f3tica.  la fusi\u00f3n gyrba posee actividad para relajar la superhelicidad positiva, pero cadece de actividad decatenasa. La actividad de gyrba mejora el crecimiento de las c\u00e9lulas, evita la escisi\u00f3n de los genes ribos\u00f3micos, pero no evita la multimerizaci\u00f3n de los minicromosomas, ni reduce la hiper-recombinaci\u00f3n mit\u00f3tica.  en conclusi\u00f3n, el exceso de superhelicidad negativa en el dna es un determinante primario de inestabilidad g\u00e9nica. En consecuencia, las tipoisomerasa i y ii act\u00faan como supresores de los procesos de recombinaci\u00f3n mit\u00f3tica relajando la superhelicidad negativa.
