Isabel <\/strong><\/h2>\n<p>El proteasoma es un complejo macromolecular responsable de la degradaci\u00f3n de la mayor parte de las prote\u00ednas intracelulares. En el presente trabajo se ha abordado el papel del proteasoma en enfermedades neurodegenerativas como proteasa en la degradaci\u00f3n de alfa-sinucle\u00edna, prote\u00edna implicada en la enfermedad de parkinson y en otras sinucleionpat\u00edas; y como autoant\u00edgeno en la esclerosis m\u00faltiple. El proteasoma 20s es responsable de la degradaci\u00f3n de alfa-sinucle\u00edna tanto in vitro como in vivo, y no se requiere previa ubiqutilaci\u00f3n. La alfa-sinucle\u00edna se comporta como un cis-degr\u00f3n y de degrada por interacci\u00f3n directa con la subunidad c8 (alfa7) del proteasoma. Las variantes de alfa-sinucle\u00edna, a30p y a53t, presentes en formas familiares de enfermedad de parkinson se comportan como alfa-sinucle\u00edna silvestre respecto a la susceptibilidad a: degradaci\u00f3n por proteasoma, fosforilaci\u00f3n por las prote\u00ednas quinsas cki, ckii, c-src y c-abl, y degradaci\u00f3n por proteasoma previa fosforilaci\u00f3n por las prote\u00ednas quinasas antes mencionadas. La vida media de alfa-sinucle\u00edna es bastante larga sugiriendo que in vivo su interacci\u00f3n con l\u00edpido de membrana y con otras prote\u00ednas previenen su degradaci\u00f3n por proteasoma. La inhibici\u00f3n de la actividad del proteasoma in vivo conduce a la agregaci\u00f3n citoplasm\u00e1tica de alfa-sinucle\u00edna, esta agregaci\u00f3n mimetiza la situaci\u00f3n que debe ocurrir en las sinucleinopat\u00edas. Todos estos resultados demuestran el papel central del proteasoma en la enfermedad de parkinson y en otras sinucleinopat\u00edas. El papel de proteasoma como autoant\u00edgeno en la esclerosis m\u00faltiple se demuestra por la presencia de anticuerpos anti-proteasoma (igg e igm), tanto en el suero (60%) como en el l\u00edquido cefalorraqu\u00eddeo (80% en pacientes seropositivos) de estos pacientes.
