{"id":15187,"date":"2018-03-09T09:02:14","date_gmt":"2018-03-09T09:02:14","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/estudios-funcionales-de-la-tirosinasa-en-el-desarrollo-de-la-retina-de-mama%c2%adferos\/"},"modified":"2018-03-09T09:02:14","modified_gmt":"2018-03-09T09:02:14","slug":"estudios-funcionales-de-la-tirosinasa-en-el-desarrollo-de-la-retina-de-mama%c2%adferos","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/ciencias-de-la-vida\/estudios-funcionales-de-la-tirosinasa-en-el-desarrollo-de-la-retina-de-mama%c2%adferos\/","title":{"rendered":"Estudios funcionales de la tirosinasa en el desarrollo de la retina de mam\u00edferos"},"content":{"rendered":"<h2>Tesis doctoral de <strong> Estela Gim\u00e9nez Caminero <\/strong><\/h2>\n<p>El objetivo principal de esta tesis es el estudio del papel de la tirosinasa en el desarrollo de la retina de rat\u00f3n.  para cumplir este objetivo se ha generado un sistema de expresi\u00f3n g\u00e9nica inducible por doxiciclina en ratones transg\u00e9nicos con el que regular, temporal y espacialmente, la expresi\u00f3n del gen de la tirosinasa de forma reversible. estudios posteriores con este sistema permitir\u00e1n evaluar si la activaci\u00f3n del gen de la tirosinasa en ratones albinos supone variaciones fenot\u00edpicas en la retina de los ratones.  por otro lado, se analiz\u00f3 el papel de la regi\u00f3n controladora de locus (lcr) del gen de la tirosinasa en el desarrollo de la retina de rat\u00f3n utilizando como herramienta ratones transg\u00e9nicos que carecen de dicha lcr. Estos estudios demostraron que dicha lcr es necesaria para el adecuado establecimiento y manteniemiento del dominio de expresi\u00f3n del gen de la tirosinasa a lo largo del desarrollo embrionario. Su ausencia provoca un retraso temporal en la iniciaci\u00f3n y acumulaci\u00f3n de pigmento en el epitelio pigmentado de la retina (rpe) durante el desarrollo.  an\u00e1lisis morfol\u00f3gicos de estos mismos ratones que carecen de la lcr del gen de la tirosinasa, en cuyas retinas conviven pigmentadas y c\u00e9lulas albinas, indicaron que las interacciones puntuales que se producen entre las c\u00e9lulas del rpe y la neuroretina de rat\u00f3n en desarrollo, que son cr\u00edticas para el desarrollo de la retina, son de tipo local y no de tipo global.  por \u00faltimo, se dilucid\u00f3 el patr\u00f3n de expresi\u00f3n del gen de la tirosinasa en etapas embrionarias mediante m\u00e9todos directos de detecci\u00f3n de expresi\u00f3n g\u00e9nica (hibridaciones in situ), demostrando que el patr\u00f3n de expresi\u00f3n del gen de la tirosinasa en rat\u00f3n se restringe al epitelio pigmentado de la retina en estadios tempranos del desarrollo embrionario.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos
