{"id":15245,"date":"2018-03-09T09:02:19","date_gmt":"2018-03-09T09:02:19","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/utilizacion-de-estrategias-de-qua%c2%admica-combinatoria-para-la-identificacion-de-moleculas-con-potencial-interes-en-biomedicina\/"},"modified":"2018-03-09T09:02:19","modified_gmt":"2018-03-09T09:02:19","slug":"utilizacion-de-estrategias-de-qua%c2%admica-combinatoria-para-la-identificacion-de-moleculas-con-potencial-interes-en-biomedicina","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/quimica\/utilizacion-de-estrategias-de-qua%c2%admica-combinatoria-para-la-identificacion-de-moleculas-con-potencial-interes-en-biomedicina\/","title":{"rendered":"Utilizaci\u00f3n de estrategias de qu\u00edmica combinatoria para la identificaci\u00f3n de mol\u00e9culas con potencial inter\u00e9s en biomedicina"},"content":{"rendered":"<h2>Tesis doctoral de <strong> Isabel Masip Masip <\/strong><\/h2>\n<p>Las t\u00e9cnicas combinatoriales han sido ampliamente utilizadas en los \u00faltimos a\u00f1os en distintos campos, especialmente en la investigaci\u00f3n biom\u00e9dica. en este trabajo se exponen los resultados obtenidos de la s\u00edntesis y posterior cribaje de distintas quimiotecas de compuestos org\u00e1nicos de peso molecular medio, frente a distintas dianas terap\u00e9uticas que comprenden \u00e1reas tan diversas como la neuroprotecci\u00f3n, la analgesia, el c\u00e1ncer, etc.  concretamente se han obtenido tres quimiotecas de peptidomim\u00e9ticos. en primer lugar se sintetiz\u00f3 una quimioteca de 2,5-piperazindionas (dkp), formada por 66 mol\u00e9culas, mediante una s\u00edntesis m\u00faltiple en paralelo en fase s\u00f3lida. Tras el cribaje de la quimioteca se hall\u00f3 una dkp que presentaba una buena actividad inhibidora de la acetilcolinesterasa por lo que se realizaron estudios de modelado molecular para establecer las principales interacciones lingado-receptor y sintetizar nuevos compuestos que potenciaran estas interacciones, con el fin de mejorar su actividad frente a dicha diana. La segunda quimioteca sintetizada const\u00f3 de 66 sales de tetraalquilamonio c\u00edclicas, mediante una secuencia en paralelo y en disoluci\u00f3n. Las etapas de purificaci\u00f3n se llevaron a cabo utilizando una resina scavenger y se agilit\u00f3 el proceso de radiaci\u00f3n por microondas.  finalmente, se sintetiz\u00f3 una quimioteca de mezclas de 5120 peptoides (tr\u00edmeros de n-alquilglicinas) mediante el formato del rastreo posicional (positional scanning). Despu\u00e9s de los ensayos biol\u00f3gicos frente a las distintas dianas, se resintetizaron los peptoides individuales que correspond\u00edan a las distintas posiciones que mostraron una mayor actividad y se volvieron a ensayar para validar los resultados. A partir de dichos resultados se han hallado peptoides con una elevada actividad analg\u00e9sica y como inhibidores del fenotipo mdr (multidrug resistance) lo que ha llevado a la realizaci\u00f3n de
