La v\u00eda de se\u00f1alizaci\u00f3n raf\/mek\/erk que conduce a la activaci\u00f3n de las prote\u00ednas erk1 y erk2 se ha revelado como un elemento clave en el control de estos procesos celulares. sin embargo, el efecto concreto de la actividad erk es dependiente del contexto celular. En este trabajo se han utilizado varias l\u00edneas celulares derivadas de leucemia mieloide. Se analiz\u00f3 el estado de activaci\u00f3n de erk en estas l\u00edneas proliferando o bajo estimulaci\u00f3n mitog\u00e9nica y en estado quiescente. Por otra parte, se estudi\u00f3 el efecto de varios agentes diferenciadores utilizados en terapia antileuc\u00e9mica sobre la actividad erk de las diversas l\u00edneas celulares. Adem\u00e1s se estudi\u00f3 el requerimiento de erk durante la proliferaci\u00f3n y diferenciaci\u00f3n mieloides utilizando un f\u00e1rmaco inhibidor de la ruta erk y un mutante inhibitorio de erk que es catal\u00edticamente inactivo, con el que se obtuvieron l\u00edneas transfectantes estables. Utilizando vectores de expresi\u00f3n que portaban distintas versiones activadas constitutivamente de mek o erk, se obtuvieron l\u00edneas trasnfectantes estables en las que se analiz\u00f3 el efecto de una activaci\u00f3n constitutiva de erk en la proliferaci\u00f3n, diferenciaci\u00f3n y supervivencia frente a est\u00edmulos apopt\u00f3picos. Se comprob\u00f3 que la proliferaci\u00f3n y diferenciaci\u00f3n de c\u00e9lulas de leucemia mieloide son independientes de la actividad erk. Sin embargo, la activaci\u00f3n constitutiva de erk gener\u00f3 una se\u00f1al de supervivencia ante distintos est\u00edmulos apopt\u00f3ticos. la localizaci\u00f3n de la actividad erk,
