A continuacion, la glucosidasa ii se solubilizo con triton x-100, se purifico a homogeneidad en ambas fuentes y se estudiaron las caracteristicas fisico-quimicas y cataliticas, no observandose ninguna diferencia significativa entre ambos lotes de ratas, salvo la estabilidad a 4 grados c. La enzima nativa es un tetramero formado por 4 subunidades iguales (mr. 106 kda). El ph optimo fue de 6,8. La energia de activacion fue de 13,54 kcal\/mol. La glucosidasa ii presenta dos centros de union para el pnp-glc: el centro activo 1 (centro de alta afinidad) y el centro 2 (centro de baja afinidad). El tipo de inhibicion -con respecto al centro 1- para la glucosa, maltosa, -d-gluconolactona, cacl2 ymgcl2 fue competitiva total, competitiva parcial, parabolica, no competitiva y no competitiva, respectivamente. La glucosa puede unirse al centro 1, la maltosa al centro 2 y la lactona a ambos centros. Tambien se describio por primera vez, el siguiente modelo de union e hidrolisis del sustrato fisiologico, donde la glucosa mas externa es liberada en el centro activo 2 de nuestro modelo, y la glucosa
