Esta tesis doctoral aborda este objetivo mediante la estrategia de vectores de secrecion de streptomyces lividans ji66. Como gen \u00abportador\u00bb se escogio el de la alfa-amilasa de s. Griseus y como gen heterologo el del factor de liberacion de la corticotropina humana (hcrf).  en una primera parte del trabajo se clono, se secuencio y se caracterizo el gen de la alfa-amilasa (amy) de streptomyces griseus en s. Lividans ji66. Como complemento a esta caracterizacion tambien se estudio la expresion del gen amy en escherichia coli como proteina de fusion con la beta-galactosidasa. Posteriormente se optimizo la expresion del gen amy mediante diferentes estrategias: incremento de su tasa transcripcional por reemplazamiento de su promotor por otros de mas eficiente expresion en s. Lividans e incremento de la tasa de secrecion de la alfa-amilasa por introduccion en su peptido \u00ableader\u00bb, a traves de tecnicas de mutagenesis \u00abin vitro\u00bb, de inserciones, sustituciones o deleciones de aminoacidos.  por ultimo se realizo la fusion traduccional entre el gen amy y el gen crf en diferentes puntos a lo largo del primero, entre los que se incluyo una fusion traduccional directa justo detras de la zona del dna que codificaba para el peptido \u00ableader\u00bb de la proteina alfa-amilasica.<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la tesis
