Nieves <\/strong><\/h2>\n<p>El objetivo principal de esta tesis ha sido el estudio del papel que determinados residuos tienen en la estructura y propiedades inmunologicas del antigeno de histocompatibilidad humano hla-a2. Se ha determinado la estructura de dos subtipos de hla-a2 denominados a 0207 y a 0208 mediante el empleo de mapas peptidicos comparativos con moleculas hla-a2 de estructura conocida.  el reducido numero de aminoacidos en que los subtipos difieren del antigeno prototipo a 0201 ha permitido identificar residuos y regiones de la molecula que participan en el reconocimiento por linfocitos t citotoxicos y anticuerpos. Este analisis tambien ha contribuido al establecimiento de dos mecanismos de diversificacion de hla-a2 y de generacion del polimorfismo en los antigenos hla de clase i.  asimismo, se ha llevado a cabo la localizacion y caracterizacion de determinantes serologicos de hla-a2 mediante el analisis de la reactividad de anticuerpos monoclonales (acms) con subtipos y mutantes de este antigeno. Los mutantes se han obtenido por mutagenesis dirigida.
