Dicha proteolisis, que produce un aumento de la afinidad del receptor, se genera de forma basal en membranas plasmaticas, hepatocitos aislados y en higado perfundido \u00abin situ\u00bb, y es incrementada por la union de insulina. Los receptores con subunidad alfa de 135 kda y de 120 kda, se translocan igualmente al interior celular. Sin embargo, el reciclamiento de estos ultimos parece estar inhibido. En hepatocitos aislados, se detecta un reciclamiento incompleto del receptor de insulina. Esto es, presumiblemente, consecuencia de la presencia en estas celulas de una alta proporcion de receptores con subunidad alfa de 120 kda. La leupeptina, al inhibir la proteolisis, permite la completa recuperacion de los receptores en la membrana plasmatica. La proteolisis de la subunidad esta inhibida \u00abin vivo\u00bb por inhibidores de proteasas presentes en el suero sanguineo, especialmente por alfa 1-antitripsina. Paralelamente, \u00abin vivo\u00bb la insulina induce en higado una redistribucion, unicamente transitoria, de sus receptores entre el interior y la superficie celular, sin modificar el contenido ni
