La base del metodo consiste en la utilizacion de las funciones topologicas de forma inversa a la convencional, es decir, en lugar de hacerlo para la prediccion de propiedades de moleculas ya existentes, se trata de emplearlas para buscar y seleccionar nuevas moleculas cuyas propiedades hayan sido prefijadas.  tras la seleccion de varias nuevas estructuras con teorica actividad analgesica y sintesis de algunas de ellas, se ha comprobado que alrededor de un 60% de los nuevos productos muestran actividad analgesica. Algunos de ellos con accion mas potente y menos toxicidad aguda (expresada como dl50) que el acido acetilsalicilico (aas) tomado como referencia.
