Paz <\/strong><\/h2>\n<p>Se ha sometido a carbamil fosfato sintetasa de e. Coli a mutag\u00e9nesis dirigida para esclarecer dos aspectos relacionados con la regulaci\u00f3n alost\u00e9rica de este enzima. Por un lado, se ha caracterizado el sitio de uni\u00f3n de los efectores nucleot\u00eddicos, ump e imp, del enzima. Se ha demostrado que ambos efectores se anclan a trav\u00e9s del grupo fosforilo a un mismo sitio en el enzima. Se ha construido un modelo de sitio de uni\u00f3n del nucle\u00f3tido efector que ha sido corroborado para el imp tras la resoluci\u00f3n de la estructura tridimensional de complejos de enzima e imp. Por otro lado, se ha establecido la independencia tanto del estado de agregaci\u00f3n del enzima con la actividad espec\u00edfica como de la olicomerizaci\u00f3n controlada por los efectores alsot\u00e9ricos en la modulaci\u00f3n de la actividad enzim\u00e1tica. Aunque ambos, oligomerizaci\u00f3n y actividad est\u00e1n controladas por los efectores alost\u00e9ricos son independientes entre s\u00ed. Para demostrar esto hemos tenido que esclarecer cual es el mecanismo de oligomerizaci\u00f3n del enzima. Cps es un (l, b) heterod\u00edmero que puede formar (l.B)2 dimeros a trav\u00e9s de interacciones entre dominios de regulaci\u00f3n de mon\u00f3meros adyacentes. La presencia de efectores positivos favorece la dimerizaci\u00f3n de los (l,b)2 dimeros a trav\u00e9s de interacciones  mutuas entre residuos pertenecientes a dominios de oligomerizaci\u00f3n de (l.B)2 d\u00edmeros adyacentes originando (l,.B)4 tetr\u00e1meros.
