Con la utilizaci\u00f3n de estas nuevas metodolog\u00edas se subsanan algunas de las limitaciones de las t\u00e9cnicas convencionales de ecolog\u00eda microbiana, as\u00ed, por ejemplo, se pueden identificar microorganismos sin necesidad de cultivarlos. En esta tesis nos planteamos la puesta a punto y aplicaci\u00f3n de t\u00e9cnicas de ecolog\u00eda molecular al estudio de la comunidad procariota de un ambiente extremo: el r\u00edo tinto.  el r\u00edo tinto es un ambiente extremo dado el bajo ph y la alta concentraci\u00f3n de metales pesados que presentan sus aguas. El bajo ph (2,3 de media) es consecuencia del metabolismo de las bacterias quimilitoaut\u00f3trofas de hierro y azufre presentes en este h\u00e1bitat. Por otro lado, la alta concentraci\u00f3n de hierro en soluci\u00f3n tampona el ph, lo que hace que sea constante a lo largo de gran parte del cauce. Para el estudio de la comunidad procariota de sus aguas se pusieron a punto tres t\u00e9cnicas de ecolog\u00eda molecular: electroforesis en geles de gradiente de desnaturalizaci\u00f3n (dgge), fluorescencia de hibridaci\u00f3n in situ (fish) y clonaje del gen del rna ribos\u00f3mico 16s.  los datos obtenidos mostraron una baja diversidad de procariotas, siendo los m\u00e1s representativos acidithiobacillus ferrooxidans, leptospirillum ferrooxidans y distintas especies del g\u00e9nero acidiphilium. Tambi\u00e9n se identificaron otros microorganismos caracter\u00edsticos de aguas \u00e1cidas de minas, como \u00abferromicrobium\u00bb y las arqueas thermoplasma acidophilum y ferroplasma acidiphilum.  estos resultados fueron comparados con los obtenidos en trabajos anteriores, realizados por nuestro grupo de investigaci\u00f3n, utilizando t\u00e9cnicas convencionales de ecolog\u00eda
