Se ha demostrado que pge1 reduce la citotoxicidad inducida in vivo por d-ga1n mediante el aumento de la expresi\u00f3n de tnf-alfa y \u00f3xido n\u00edtrico sintasa inducible (inos) en hepatocitos.  el presente estudio se dise\u00f1\u00f3 para determinar el papel del tnf-alfa en la apoptosis y necrosis inducida por d-ga1n y su protecci\u00f3n por pge1 en el cultivo primario de hepatocitos de rata en co-cultivo con c\u00e9lulas de kupffer. Los hepatocitos y las c\u00e9lulas de kupffer se aislaron de ratas macho mediante la perfusi\u00f3n con colagenasa y la purificaci\u00f3n por centrifugaci\u00f3n diferencial y elutriaci\u00f3n, respectivamente. Los hepatocitos se co-cultivaron con las c\u00e9lulas de kupffer en una relaci\u00f3n de 3,5:1 con la utilizaci\u00f3n de insertos de cultivo.  para determinar el papel del tnf-alfa, se a\u00f1adieron anticuerpos anti-tnf-alfa (0,3 mg\/ml) 2h antes que la pge1 \u00f3 4h antes que la d-ga1n. La apoptosis celular se evalu\u00f3 con la aparici\u00f3n de fragmentaci\u00f3n del dna, y la necrosis mediante la liberaci\u00f3n de lactato deshidrogenasa (ldh) al medio de cultivo. la producci\u00f3n de \u00f3xido n\u00edtrico (on) se determin\u00f3 por la cuantificaci\u00f3n de los productos finales, nitrito+nitrato (nox), mediante el reactivo de griess. Tnf-a, il-1a, il-6, e ifn-g fueron cuantificados por elisa en el medio de cultivo. La lipoperoxidaci\u00f3n se determin\u00f3 mediante la valoraci\u00f3n de malondialdeh\u00eddo (mda) en el medio de cultivo. Se observ\u00f3 que la inducci\u00f3n de apoptosis y necrosis por d-ga1n en los hepatocitos se asoci\u00f3 con un aumento de on, tnf-alfa y mda en el medio de cultivo. La protecci\u00f3n por pge1 de la apoptosis por d-ga1n se asocia con una reducci\u00f3n de la concentraci\u00f3n de on en el medio de cultivo. El co-cultivo de los hepatocitos con c\u00e9lulas de kupffer exacerba la necrosis y apoptosis inducida por d-ga1n.
