En la presente tesis se estudia el proceso de maduraci\u00f3n de c\u00e9lulas dendr\u00edticas humanas derivadas de monocito in vitro identific\u00e1ndose:  1,- un aumento en la expresi\u00f3n de integrinas cd49d (alfa4\/beta1 y alfa4\/beta7) durante la maduraci\u00f3n, que permite considerar a cd49d como un nuevo marcador de la maduraci\u00f3n de c\u00e9lulas dendr\u00edticas humanas derivadas de monocito in vitro.  2,- un efecto regulador negativo de la v\u00eda se\u00f1alizaci\u00f3n de mek-erk sobre la maduraci\u00f3n fenot\u00edpica y funcional, y regulador positivo de p38mapk en la misma. En consecuencia, las rutas de mek-erk y p38mapk tienen efectos opuestos en la maduraci\u00f3n de c\u00e9lulas dendr\u00edticas derivadas de monocito.  tambi\u00e9n se estudi\u00f3 la regulaci\u00f3n transcripcional de la integrina leucocitaria cd11a, que se expresa exclusivamente en leucocitos y su nivel de expresi\u00f3n var\u00edan seg\u00fan el estado de diferenciaci\u00f3n celular. En la regi\u00f3n reguladora proximal del gen de cd11a se identificaron dos secuencias solapantes de reconocimiento de los factores de transcripci\u00f3n aml\/cbfbeta (aml-110) y c\/ebp (c\/ebp-100). En c\u00e9lulas de origen linfoide el promotor se encuentra regulado por la familia de factores de transcripci\u00f3n aml, mientras que en c\u00e9lulas de origen mieloide la regulaci\u00f3n depende de los factores de transcripci\u00f3n c\/ebp. Es decir, la regulaci\u00f3n es linaje dependiente.  por \u00faltimo, la mol\u00e9cula pre-leuc\u00e9mica aml-1\/eto generada por la traslocaci\u00f3n cromosomal t(8;21) en c\u00e9lulas de leucemia mieloide aguda reprime la transcripci\u00f3n de cd11a a trav\u00e9s del reconocimiento de aml-110, lo que permite explicar la baja\/nula expresi\u00f3n de cd11a en c\u00e9lulas de leucemia mieloide
