Inmaculada Monle\u00f3n Brau <\/strong><\/h2>\n<p>En el presente trabajo se ha estudiado la muerte celular inducida por activaci\u00f3n (aicd) en la leucemia t humana jurkat, subl\u00edneas derivadas (j.Rt3-t3.5 jhp y jws) y blastos t humanos normales. Se ha comprobado la implicaci\u00f3n de fasl y apo2l en el proceso de aicd inducido por pha o por anti-cd3, adem\u00e1s los resultados obtenidos sugieren la participaci\u00f3n de alg\u00fan mensajero de muerte adicional, todav\u00eda sin caracterizar. Asimismo, se ha estudiado la aicd en estas c\u00e9lulas a trav\u00e9s de glicoprote\u00ednas de superficie distintas del tcr, en concreto cd59. Fasl y apo2l son secretados al sobrenadante durante el proceso de aicd en las c\u00e9lulas jurkat o blastos t humanos en su forma intacta, no proteolizada, asociadas a una fracci\u00f3n particulada y ultracentrifugable. Se ha caracterizado la localizaci\u00f3n intracelular de estas prote\u00ednas, comprobando que fasl y apo2l se encuentran mayoritariamente almacenados en cuerpos multivesiculares (mvbs) del citoplasma de estas c\u00e9lulas con un tama\u00f1o entre 400 y 600 nm y su secreci\u00f3n est\u00e1 asociada con microves\u00edculas. La ligaci\u00f3n de cd59 induce la secreci\u00f3n preferente de apo2l, sobre todo en los blastos t normales.  se ha caracterizado que las subl\u00edneas hiperproliferativas derivadas de jurkat generadas previamente en nuestro laboratorio jhp y jws son resistentes a la aicd. Esto se correlaciona con la ausencia de expresi\u00f3n en su membrana plasm\u00e1tica tanto de mol\u00e9culas de activaci\u00f3n propias de las c\u00e9lulas t (cd3, cd2, cd59), como del receptor proapopt\u00f3tico fas\/cd95. La importancia del sistema fas\/fasl en la aicd se confirma por el hecho de que las subl\u00edneas han perdido tambi\u00e9n la expresi\u00f3n intracelular de fasl.  se ha analizado el mecanismo por el que fas se acumula en el interior celular de las subl\u00edneas proliferativas derivadas de jurkat. Debido a un fallo en la ruta secretora, fas es conducido hacia compartimentos lisomonales para su degradaci\u00f3n.
