En el presente trabajo, se ha estudiado la regulaci\u00f3n de los genes tap1 y lmp2. Estos genes codifican prote\u00ednas cuyas funciones son vitales para la presentaci\u00f3n antig\u00e9nica de prote\u00ednas end\u00f3genas a trav\u00e9s del mhc de clase l. La prote\u00edna lmp2 forma parte de un complejo multiproteico conocido como proteosoma, cuya funci\u00f3n es la degradaci\u00f3n de prote\u00ednas citos\u00f3licas dando lugar a la formaci\u00f3n de p\u00e9ptidos. La prote\u00edna tap1 est\u00e1 implicada en el transporte de los p\u00e9ptidos al interior ret\u00edculo endoplasm\u00e1tico, para su posterior carga en las mol\u00e9culas de histocompatibilidad de clase l. Ambas prote\u00ednas son codificadas por genes que comparten una misma regi\u00f3n promotora en sentidos opuestos.  los resultados confirman que la inducci\u00f3n de la expresi\u00f3n de los genes murinos tap1 y lmps mediada por el ifn-gamma tiene lugar en c\u00e9lulas de diferentes linajes y ocurre entre una y tres horas post-activaci\u00f3n. El an\u00e1lisis de otros est\u00edmulos inductores de la expresi\u00f3n de tap1 y lmp2 demostr\u00f3 patrones de expresi\u00f3n diferentes del obtenido con el ifn-gamma. El lps induce la expresi\u00f3n de tap1 y lmp2 a las seis horas de activaci\u00f3n y el tnf-alfa a las 24 horas. En nuestros experimentos, observamos, en macr\u00f3fagos derivados de m\u00e9dulas \u00f3sea mantenidos en medio de cultivo en ausencia de suero (situaci\u00f3n de estr\u00e9s), una inducci\u00f3n de la expresi\u00f3n de los genes tap1 y lmp2. Esta inducci\u00f3n tiene lugar aproximadamente a las 24 horas del est\u00edmulo, siguiendo un patr\u00f3n de inducci\u00f3n semejante al observado tras la activaci\u00f3n con el tnf-alfa.  utilizando cicloheximida, un inhibidor de la s\u00edntesis proteica, se comprob\u00f3 que la inducci\u00f3n de tap1 y lmp2 por el ifn-gamma no es dependiente de la s\u00edntesis proteica. Los tr\u00e1nscritos de arn son altamente estables, ya que sus niveles no decaen de forma r\u00e1pida en presencia de actino<\/p>\n<p>&nbsp;<\/p>\n<h3>Datos acad\u00e9micos de la tesis doctoral
