De <\/strong><\/h2>\n<p>Cot quinasa, una mapkkk involucrada en la activaci\u00f3n de la c\u00e9lula t, regula la expresi\u00f3n del gen cox-2 (ciclooxigenasa-2) en una l\u00ednea celular humana de lifoncitos t (jurkat). La inducci\u00f3n producidas por cot quinasa en la actividad promotora de cox-2, se produce mediante la activaci\u00f3n de los elementos distal (-105\/97) y pr\u00f3ximal (-76\/-61) del factor de transcripci\u00f3n nuclear de la c\u00e9lula t (nfat) de este promotor. Adem\u00e1s, cot quinasa incrementa la transactivaci\u00f3n mediada por nfat. A diferencia de la estimulaci\u00f3n de la actividad transactivadora de nfat y del promotor de cox-2 por esteres del forbol e ion\u00f3foro de calcio que es inhibible por la droga inmunosupresora ciclosporina a (csa), el incremento en la actividad promotora de cox-2 y de la transactivaci\u00f3n mediada por nfat inducido por la actividad cot quinasa es independiente de csa.  hemos evaluado la capacidad que posee el metamizol de inhibir las actividades enzim\u00e1ticas cox-1 y cox-2 usando diferentes fuentes de cox. El metamizol inhibe la actividad enzim\u00e1tica cox-1 y cox-2 con una ic50 de aproximadamente 150 ug\/ml en enzimas purificadas, siendo similar a la ic50 obtenida al inhibir con metamizol extractos de macr\u00f3fagos murinos previamente inducidos con lps. Sin embargo los resultados obtenidos en c\u00e9lulas intactas son diferentes: la ic50 del metamizol para cox-1 en c\u00e9lulas baec (c\u00e9lulas endoteliales de aorta de oveja) y en plaquetas humanas son 1730 +\/-150 ug\/ml y 486+\/-56 ug\/ml respectivamente. La ic50 del metamizol sobre cox-2 en macr\u00f3fagos murinos y en pbls humanos (linfocitos de sangre perif\u00e9rica), ambos activados previamente con lps son 12+\/-1.8 ug\/ml y 21+\/-2.9 ug\/ml respectivamente. estos datos nos indican que al menos parte de las acciones farmacol\u00f3gicas del metamizol podr\u00edan estar mediadas por la regulaci\u00f3n de la actividad cox-2.
