En este estudio evaluamos la capacidad de un adenovirus portador de los genes de la interleuquina-12 (il-12) murina (adcmvil-12) para erradicar n\u00f3dulos tumorales en dos modelos diferentes de hepatocarcinoma ortot\u00f3pico desarrollados en ratas buffalo. La inyecci\u00f3n intratumoral de adcmvil-12 en animales portadores de un \u00fanico n\u00f3dulo implantado en el h\u00edgado ha demostrado inhibir significativamente el crecimiento tumoral con un claro patr\u00f3n dosis-dependiente. El 50% de los animales recibieron una dosis de 5&#215;10(9) pfu, mostraron regresiones tumorales completas dos semanas despu\u00e9s del tratamiento. En animales con dos n\u00f3dulos tumorales independientes generados en el l\u00f3bulo mayor izquierdo del h\u00edgado, la inyecci\u00f3n de 5&#215;10(9) pfu de adcmvil-12 en una de las dos lesiones mostr\u00f3 un 50% de remisiones completas tanto en los tumores tratados como en los n\u00f3dulos adyacentes.  estos datos quedan confirmados con una supervivencia del 66,7% de los animales incluidos en este grupo. Por otra parte, las ratas que quedaron libres de tumores despu\u00e9s de haber recibido el tratamiento con adcmvil-12 mostraron una protecci\u00f3n frente a una nueva inyecci\u00f3n de la misma l\u00ednea celular (mca-rg7777) utilizada para inducir los tumores iniciales. Adem\u00e1s, encontramos que los animales tratados con adcmvil-12 producen niveles s\u00e9ricos elevados de il-12 el primer d\u00eda despu\u00e9s del tratamiento y altos valores de ifn-y los d\u00edas 1 y 7 despu\u00e9s de la administraci\u00f3n de este vector. En este sentido, encontramos que la activaci\u00f3n de las c\u00e9lulas nk y los linfocitos t cd8+, as\u00ed como la inhibici\u00f3n de la angiog\u00e9nesis son los mecanismos antitumorales que es capaz de activar el adcmvil-12.
