{"id":17089,"date":"2018-03-09T09:05:00","date_gmt":"2018-03-09T09:05:00","guid":{"rendered":"https:\/\/www.deberes.net\/tesis\/sin-categoria\/studies-on-uev-a-new-regulator-of-polyubiquitination-functional-aspects-and-genomic-analysis\/"},"modified":"2018-03-09T09:05:00","modified_gmt":"2018-03-09T09:05:00","slug":"studies-on-uev-a-new-regulator-of-polyubiquitination-functional-aspects-and-genomic-analysis","status":"publish","type":"post","link":"https:\/\/www.deberes.net\/tesis\/quimica\/studies-on-uev-a-new-regulator-of-polyubiquitination-functional-aspects-and-genomic-analysis\/","title":{"rendered":"Studies on uev, a new regulator of polyubiquitination: functional aspects and genomic analysis"},"content":{"rendered":"<h2>Tesis doctoral de <strong> Noureddine Loukili <\/strong><\/h2>\n<p>Nuestros objetivos en esta tesis han sido:  1,- estudiar la funci\u00f3n de la prote\u00edna uev1, un nuevo regulador de poliubiquitinaci\u00f3n encontrado en la b\u00fasqueda de nuevos genes implicados en el control de la diferenciaci\u00f3n y la proliferaci\u00f3n celular.  2,- analizar la arquitectura gen\u00f3mica del gen para uev1.  3,- establecer un an\u00e1lisis comparativo de estos genes y prote\u00ednas, incluyendo estudios filogen\u00e9ticos y evolutivos.  mediante diferencial display (rap) con rna de una l\u00ednea celular th29 m6 de c\u00e1ncer de colon hemos identificado una nueva familia de prote\u00ednas altamente conservadas en todos los eucariotas que son variantes de los enzimas de conjugaci\u00f3n de ubiquitinas y que no posen el centro catal\u00edtico intr\u00ednseco de estas. A estas prote\u00ednas las hemos denominado uev. Hemos demostrado que la prote\u00edna uev por si sola no promueve la conjugaci\u00f3n de la ubiquitina.  hemos encontrado al menos tres isoformas para este transcrito y que este transcrito mediante secuenciaci\u00f3n y comparaci\u00f3n con datos genomicos hemos deducido que estas 3 isoformas se deben a un splicing alternativo.  mediante fish utilizando clones pac que contienen el gen uev1 hemos podido localizar este gen uev1 en el cromosoma 20q13.2.  desde el punto de vista funcional hemos observado mediante citometria de flujo que c\u00e9lulas m6 transfectadas para una expresi\u00f3n constitutiva del uev1 presentan una mayor acumulaci\u00f3n en fase s y g2m.  adem\u00e1s de la alta proporci\u00f3n de c\u00e9lulas en fase s y g2m que en el control hemos observado una acumulaci\u00f3n de c\u00e9lulas multinucleadas y que aumenta en proporci\u00f3n con el tiempo de cultivo.  tambi\u00e9n hemos detectado un 3% de c\u00e9lulas apopt\u00f3ticas que no existen en el control.  hemos visto que el menos parte de este fenotipo se asocia a una inhibici\u00f3n de la actividad de la kinasa mitotica cdk1.  hemos demostrado la presencia de un nuevo gen, que hemos llamado
